To run the 2017.16 method (RINTDF) on the ANKOMTDF Dietary Fiber Analyzer, you will run method 2009.01 with the modifications detailed below.

Before running the 2017.16 method, make the following modifications to the 2009.01 method:
1) Shorten the digestion time.
2) Modify the enzyme dilutions.

Before running the 2017.16 method, make the following modifications to the 2009.01 method:
1) Shorten the digestion time.
On the “Select a Function” screen on the instrument’s Touch Screen Display, press the “Diagnostics” button. Next, press “Digestions Times.” This screen will allow you to adjust the times. For the 2009.01 method, change the 16 hour digestion to 4 hours. The new digestion time will remain until you change the time again.
2) Modify the enzyme dilutions.
The target activity is 4000 U (units of activity) PAA (Porcine Pancreatic a-Amylase)/sample, 1700 U AMG/sample and 35 U Protease/sample. The first enzyme position (PAA and AMG combined) will deliver two mL per sample and the 2nd position (Protease) will deliver 1mL per sample.
Assuming you are using the ANKOM enzymes you can dilute 1.06g of the Porcine Pancreatic a-Amylase (TDF130) to 20 ml with Sodium Maleate buffer (properly prepared per the method). Separately dilute 10.42mL of AMG (TDF84 or TDF85) to 20 mL with Sodium Maleate buffer (9.58mL). Combine the two for a total of 40ml. You will use this mixture directly in the first enzyme position (PAA and AMG combined).
The Protease enzyme has a target of 35 U/sample and goes in the 2nd position on the instrument. The Protease dilution will be the same as used for the 991.43, 2009.01 and 2011.25 methods. For ANKOM Protease enzymes TDF82 or TDF83, dilute 5mL enzyme to 25mL with distilled water.
Use the same Trizma Base solution in position 3 on the instrument as is specified for the 2009.01 method.
If you use non-ANKOM enzymes, you must make sure that the dilutions produce the Units of Activity as noted above (4000 U PAA/sample, 1700 U AMG/sample and 35 U Protease/sample) during the digestion, keeping in mind that the instrument delivers 2 mL per sample from the first enzyme position and 1mL per sample from the 2nd position.
